Prostate

Supplied 34bE12

A section of prostatic tissue immuno-stained with 34bE12 which is a monoclonal mouse antibody against human high molecular weight cytokeratin. The antibody labels normal squamous, ductal and other complex epithelia. It reacts with benign small-acinous lesions of the prostate. Consistent positive results are found with squamous cell carcinomas, ductal carcinomas, transitional cell carcinomas, thymomas, and epithelioid mesotheliomas. Adenocarcinomas are variably positive.

 

In-house 34bE12

The section from the same tissue block stained with the in-house 34bE12. There is not significant difference between the supplied and in-house antibodies. The best results for both demonstrations are obtained by peroxidase blocking for 8 min., 2 rounds of microwave irradiation for 13 min. and 8 min., and immuno-stained with 1:30 diluted primary antibody for 45 min. at room temperature.


Prostate Specific Antigen

A section of prostatic tissue infiltrated by adenocarcinoma, which occupies more than 90% of the tissue area. Immunohistochemically, the tumour cells express prostate specific antigen (PSA). The PSA demonstration is well performed with good signal to background ratio; i.e. high target intensity versus clear background. The best results are obtained by peroxidase blocking for 10 min., pressure cooking for 2.5 min., and immuno-stained with 1:1000 diluted primary antibody for 32 min. at 37 °C.
